Comparison of mesenchymal stromal cells from peritoneal dialysis effluent with those from umbilical cords: characteristics and therapeutic effects on chronic peritoneal dialysis in uremic rats

Abstract Background A long-term of peritoneal dialysis (PD) using a hypertonic PD solution (PDS) leads to patient’s membrane (PM) injury, resulting in ultrafiltration failure (UFF) and drop-out. Our previous study shows that effluent-derived mesenchymal stromal cells (pMSCs) prevent the PM injury normal rats after repeated exposure cavity PDS. This was designed compare cytoprotection between pMSCs umbilical cord-derived MSCs (UC-MSCs) treatment both kidney uremic with chronic PD. Methods 5/6 nephrectomized (5/6Nx) Sprague Dawley were intraperitoneally (IP) injected Dianeal (4.25% dextrose, 10 mL/rat/day) treated or cord (UC)-MSCs (approximately 2 × 6 /rat/week, IP). Ultrafiltration determined by IP injection 30 mL dextrose) 1.5-h dewell time, serum creatinine (SCr) blood urea nitrogen (BUN). The structure kidneys assessed histology. Gene expression examined quantitative reverse transcription PCR, protein levels flow cytometric Western blot analyses. Results We showed slight difference morphology UC-MSCs plastic dishes, significantly higher stemness-related genes ( NANOG , OCT4 SOX2 CCNA2 RAD21 EXO1 ) surface markers (CD29, CD44, CD90 CD105) than those pMSCs, but no differentiation chondrocytes, osteocytes adipocytes. pMSC more effective protection MP remnant 5/6Nx from PDS-induced which associated resistance toxins culture, reduction mesothelial cell death secretome response PDS exposure. similarly inactivated NOS2 activated THP1 cells. Conclusions As compared UC-MSCs, may potently this rat model PD, suggesting autotransplantation ex vivo-expanded become promising therapy for UFF deterioration function patients.
